Si @silicon

While we're thinking about science, who knows shit about microscopes?

I would love to know of a microscope that can simultaneously 1) image high enough resolution to resolve FISH spots and 2) be able to scan an entire organ and longshot 3) do it in 3D (not slices)?

:D

Okay I know this is probably a pipe dream but it WOULD BE AWESOME and my project hella needs it.

@silicon How big is the organ you want to look at? smFISH has been done in whole-mount zebrafish embryos, check out nature.com/articles/srep08571 (open access)

@quokka That paper is super cool! Mine's around 1cm x .5cm x .5 cm (VERY roughly).

@silicon hmmm that might be a bit large to image in one go, for smFISH anyway. I don't know if anyone has done FISH on something like a SPIM or lattice light-sheet microscope (if you don't require super-high or single-molecule spatial resolution).

Here's another paper using FISH with expansion microscopy: nature.com/articles/nmeth.3899

You might also need to look at clearing the organ with something like CLARITY although again I don't know if that has been used with FISH as well.
